The mitochondrion undergoes mitoflashes, shown as flaming peaks for heat production. As the powerhouse of the cell, mitochondria produce ATP, the energy currency of life, through respiratory chain complexes that can form a megacomplex (shown in the bubble), whose structure is being resolved by Zhang and his colleagues. According to the conformational change of megacomplex, it is concluded that respiratory chain complex I, complex III, and complex IV work in a synergistical manner, in which the activation state of complex I can influence the Rieske domain movement of complex III and the swing range of complex IV. Mitochondria also generate elementary electrochemical excitations in the form of mitoflashes, which represent transient uncoupling of mitochondrial respiration. Chen and her colleagues show a close link between mitoflashes and heat production. They find that when the body is challenged with environmental cold, mitoflashes are extensively activated in brown adipocytes, a specialised cell type for adaptive thermogenesis. As stimulation of brown adipocyte thermogenesis increases systematic energy expenditure, understanding mitoflash signalling in heat production will open up avenues to combat obesity and its associated metabolic disorders.
